چکیده

Background Particulate air pollution can induce epigenetic changes and regulate gene expression relevant to the pathophysiology of asthma and allergic diseases. Recently, epidemiologic data suggests that there are observable acute effects of air pollution on peripheral blood DNA methylation levels of genomewide Alu and LINE-1 repeat elements, as well as certain genes involved in oxidative stress response and innate immunity. In this study, we hypothesized that in a controlled exposure setting, diesel exhaust (as a model of

Effects of controlled diesel exhaust exposure on apoptosis and proliferation markers in bronchial epithelium – an in vivo bronchoscopy study on asthmatics, rhinitics and healthy subjects

BACKGROUND Epidemiological evidence demonstrates that exposure to traffic-derived pollution worsens respiratory symptoms in asthmatics, but controlled human exposure studies have failed to provide a mechanism for this effect. Here we investigated whether diesel exhaust (DE) would induce apoptosis or proliferation in the bronchial epithelium in vivo and thus contribute to respiratory symptoms. ...

Combined inhaled diesel exhaust particles and allergen exposure alter methylation of T helper genes and IgE production in vivo.

Changes in methylation of CpG sites at the interleukin (IL)-4 and interferon (IFN)-gamma promoters are associated with T helper (Th) 2 polarization in vitro. No previous studies have examined whether air pollution or allergen exposure alters methylation of these two genes in vivo. We hypothesized that diesel exhaust particles (DEP) would induce hypermethylation of the IFN-gamma promoter and hyp...

Diesel exhaust enhances airway responsiveness in asthmatic subjects.

Particulate matter (PM) pollution has been associated with negative health effects, including exacerbations of asthma following exposure to PM peaks. The aim of the present study was to investigate the effects of short-term exposure to diesel exhaust (DE) in asthmatics, by specifically addressing the effects on airway hyperresponsiveness, lung function and airway inflammation. Fourteen nonsmoki...

Health effects of diesel exhaust emissions.

Epidemiological studies have demonstrated an association between different levels of air pollution and various health outcomes including mortality, exacerbation of asthma, chronic bronchitis, respiratory tract infections, ischaemic heart disease and stroke. Of the motor vehicle generated air pollutants, diesel exhaust particles account for a highly significant percentage of the particles emitte...

Flow cytometry to identify leukocyte sub-populations in blood and induced sputum in asthmatic and healthy volunteers exposed to diesel exhaust

Methods Mild asthmatics and normal controls were recruited as study subjects. This crossover study was double-blinded, randomized and counter-balanced to the order of three conditions: diesel exhaust with anti-oxidant, diesel exhaust with placebo, or filtered air with placebo. The subjects were exposed to either filtered air or diesel exhaust (300 ug PM2.5/m ) in a state-of-the-art diesel exhau...
